Arrestins (arr) are multifunctional cytosolic adaptors that bind to active and phosphorylated G protein-coupled receptors (GPCRs) via a highly versatile interface. stop protein signaling trigger other pathways. Recently, 3D structures of arr–GPCR complexes have been solved, which provide bulk structural information for understanding the mechanism arr recruitment activation. However, many questions about functional consequences details dynamics interaction remain open. A wealth key determinants their relevance, dynamic insights into process binding outcomes different modes provided by series biochemical methods we review here.
